现代研究表明,人体摄入的蛋白主要是以小肽的形式在肠道直接吸收。氨基酸和肽的吸收转移机制不同,肽的吸收效率更高且不易饱和,例如二肽和三肽的吸收速度 高于同一组成的氨基酸。经酶解后得到食用肽类制品具有易吸收、抗氧化、抗疲劳、降血脂、降血压、增强免疫力等一系列生理功能,食源性肽类已成为国际上最具 发展潜力的功能性原料。本课题以工业化水平的食源性低聚肽(大豆低聚肽、海洋系列低聚肽、白蛋白肽、乳清蛋白低聚肽、玉米肽等)为研究样品,采用柱色谱及 高效液相色谱法对其内含的混合肽成份进行分离、测序,确定不同蛋白来源肽原料的主要肽组份与结构;对照功能肽数据库中已知的功能结构确定功能因子并以抗氧 化及降血压体系确定活性肽的构效关系,研究其作用机理;在已进行的大量动物体内功能实验及临床实验基础上,进行食源性肽类原料体外功能评价体系的试验工 作,建立起适合产业需求的肽类原料快速功能评价体系。

Identification of industrialized food-derived oligopeptides and establishment of in vitro and in vivo activity assay system

Modern research shows that dietary proteins are absorbed directly mainly in the form of small peptides in the intestinal tract. Compared to amino acids, peptides have higher absorption efficiency and are difficult to saturate due to the differences in the absorption system. For example, dipeptides and tripeptides are absorbed more rapidly than free amino acid mixtures in the intestinal tract. Peptides derived from food proteins by enzymatic hydrolysis exhibit many physiological effects, such as antioxidant, antifatigue, antihyperlipidemic, antihypertensive, immunomodulatory activity, and enhancement of absorption. Food-derived peptides have become one of the most potential functional ingredients in the world. In this research project,industrialized food-derived oligopeptides derived from various protein sources such as soy, marine fish, egg albumin, whey, corn, were used as the samples. For the purpose of peptide sequencing, the oligopeptides were first isolated by column chromatography and high performance liquid chromatography, and then the molecular mass and amino acid sequence were identified by mass spectrometer. Afterwards, the bioactive peptides identified among these peptides were compared with the bioactive peptide database; the relationship between antioxidant or antihypertensive behavior and structure as well as the mechanism of action were investigated. Based on plentiful animal experiments and clinical trials, the analysis of in vitro activities of food-derived peptides was carried on, and a rapid activity assay system of peptide materials that could meet the demands of the industry was established.
